Dermal-epidermal junction (DEJ) stimulation

The DEJ is an important structural component of skin, providing a critical link between the basement membrane and keratinocytes. Its disruption is responsible for aesthetic skin care sub-epidermal blistering dermatoses including bullous pemphigoid, dermatitis herpetiformis and epidermolysis bullosa acquisita. In these disorders, auto-antibodies target components of the basement membrane at the DEJ leading to blister formation. Immunohistochemistry has shown that GzmB accumulates at the DEJ during blister formation1.

We have recently demonstrated that topical treatment with palmitoyl-arginyl-gly-asp (PAL-RGD) improves wrinkles, skin elasticity and dermal density in human HaCaT keratinocytes1. PAL-RGD induces collagen XVII expression in keratinocytes, which is a key component of the DEJ. This protein is involved in epidermo-dermal adhesion, binding laminin-332 and collagen IV to form a heterogenous bridge between the stroma and the ectoderm. It also binds fibronectin and acts as a transmembrane anchor for cytoplasmic proteins, linking ectoderm and dermis.

During the aging process, the protein components of the DEJ decrease. The result is a loss of the DEJ membrane thickness, flattening of the rete ridges and disorganization of lamina lucida and lamina aesthetic skin care shop densa. The results of the senescence studies indicate that MB at nanomolar concentration is capable to scavenge free radicals in dermal fibroblasts and protect cells against oxidative damage by inhibiting MMP degradation of the ECM. Moreover, MB increases the expression of ECM proteins in 3D-reconstructed human skin models. The expression of ECM components such as elastin and Col2A1 was significantly up-regulated by MB treatment compared to controls.

Additionally, the MB-induced increase in skin hydration and thickness is related to the up-regulation of ECM protein expression. Specifically, MB significantly increased the expression of elastin in skin tissue derived from two old dermal fibroblast lines (3-OM and 4-OF). The results suggest that BM can slow down the aging process by reducing skin damage caused by ROS and reversing the senescence phenotypes.

Biotinylated peptides

In the present study, a biotinylated peptides complex stimulating skin basement membrane proteins expression was developed and its anti-wrinkle effect was investigated ex vivo. Moreover, the effect of the peptides complex on wrinkle reduction was assessed in a clinical test with human volunteers. The results showed that the peptides complex has both ex vivo and in vitro anti-wrinkle activity, which suggests that it can be used as an anti-aging cosmetic ingredient.

To develop a biotinylated ligand, a peptide with a carboxylic acid group was selected. It was then conjugated with a biotin molecule via the enzymatic synthesis procedure. The resulting biotinylated peptide was analyzed by LC-MS/MS. The search parameters were a trypsin enzyme with up to two missed cleavages and a precursor mass tolerance of 10 ppm. The mass spectrum was normalized, and the peptides were identified using the precursor ion quantifier node. Carboxylation of cysteine, oxidation of methionine, and biotinylation of lysine were identified as variable modifications.

To assess the effects of the biotinylated peptides on BM protein expression in epidermal cells, a soluble form of the pentapeptide YIGSR (tyrosine-isoleucine-glycine-serine-arginine) was synthesized. This peptide was biotinylated with a biotin-containing reagent, and its binding ability to BM proteins was tested in cultured human epidermal keratinocytes. The peptide significantly increased the expression of laminin 522 and nidogen in high (200 ppm) concentrations. It also stimulated collagen synthesis in cultured dermal fibroblasts.
